ABSTRACT

OBJECTIVE@#To evaluate the therapeutic effects of acupoint autohemotherapy (A-AHT) on 1-chloro-2,4-dinitrobenzene (DNCB)-induced atopic dermatitis (AD) in mice focusing on regulating T helper 1/T helper 2 (Th1/Th2) immune responses.@*METHODS@#Thirty BALB/c mice were divided into 5 groups by a random number table, including normal control (NC), AD model (AD), A-AHT, sham A-AHT (sA-AHT), and acupoint injection of normal saline (A-NS) groups, 6 mice per group. Mice were challenged by DNCB for the establishment of experimental AD model. On the 8th day, except for the NC and AD groups, the mice in the other groups received management once every other day for a total of 28 days. For the A-AHT and sA-AHT groups, 0.05 mL of autologous whole blood (AWB) was injected into bilateral Zusanli (ST 36) and Quchi (LI 11) and sham-acupoints (5 mm lateral to ST 36 and LI 11), respectively. The A-NS group was administrated with 0.05 mL of normal saline by acupoint injection into ST 36 and LI 11. Dermatitis severity for dorsal skin of mice was determined using the Severity Scoring of Atopic Dermatitis (SCORAD) every week. The total immunoglobulin E (IgE), interleukin-4 (IL-4), and interferon-gamma (IFN-γ) cytokine levels in serum were examined by enzyme-linked immunosorbent assay (ELISA). Spleen Th1/Th2 expression were analyzed via flow cytometry and immunohistochemical assay was used to detect T-box expressed in T cell (T-bet) and GATA-binding protein 3 (GATA3) expressions in skin lesions of mice.@*RESULTS@#Compared with the AD group, both A-AHT and sA-AHT reduced the SCORAD index and serum IgE level (P<0.05 or P<0.01); A-AHT, sA-AHT and A-NS down-regulated serum IL-4 level and upregulated IFN-γ/IL-4 ratio (P<0.05 or P<0.01); A-AHT regulated the Th1/Th2 shift specifically and increased the related transcription factors such as T-bet expression and T-bet/GATA3 ratio (P<0.05).@*CONCLUSION@#A-AHT showed significant effectiveness on the AD model mice, through regulating Th1/Th2 immune responses.

ABSTRACT

Degradation of 2,4,6-trinitrotoluene (TNT), a nitroaromatic explosive found in the soil and ground water, was investigated using Pseudomonas aeruginosa in in vitro experiments. Biodegradable abilitiy of this bacteria was performed with 50 and 75 mg L−1 TNT concentrations in a defined liquid medium for 96 h time period. Treatment of TNT in supernatant samples taken at 0, 6, 12, 24, 48, 72 and 96 h from agitated vessels was followed by reverse-phase high-performance liquid chromatography (HPLC). In cultures supplemented with 50 and 75 mgL−1 TNT, after 96 h of incubation 46% and 59% reduction were detected respectively. Two metabolites as degradation intermediates with nitrite release into the medium, 2,4-dinitrotoluene (2,4-DNT) and 4-aminodinitrotoluene (4-ADNT), were elucidated by thin layer chromatography (TLC) and gas chromatography-mass spectrometry (GC-MS). These findings clearly indicate that Pseudomonas aeruginosa can be used in bioremediation of TNT contaminated sites.

ABSTRACT

Propõe-se, neste trabalho, avaliar a quantidade de formaldeído deixado no interior de canais radiculares, quando eles são secos com cones de papel absorvente mantidos em ambiente com pastilhas de formaldeído. Cones de papel tratados com 2,4-Dinitrofenil-hidrazina foram utilizados para a captação dessa substância, e, em seguida, foram submetidos à análise por Cromatografia Líquida de Alta Eficiência (CLAE) para a quantificação desse formaldeído residual. Verificou-se que cones de papel expostos ao formaldeído deixam resíduos dessa substância no interior dos condutos radiculares, fato preocupante, uma vez que a referida substância tem potencial carcinogênico.

ABSTRACT

Three main enzymes, responsible for bioconversion of 1,3 dinitrobenzene (m-DNB) by Micrococcus colpogenes MCM B 410, were isolated from the sonicated cell mass, grown in presence of m-DNB in a synthetic medium, for 7 days. The soluble proteins were separated by differential precipitation and also separated by native PAGE. Each fraction obtained from both the protocols, was tested for nitro aryl reductase, aryl mono oxygenase and resorcinol 2,3 dioxygenase. The apparent molecular weight of the proteins were nitro aryl reductase (30 kDa), aryl mono oxygenase (110 kDa) and resorcinol 2,3 di oxygenase (65 kDa).

ABSTRACT

Micrococcus colpogenes MCM B 410, indigenous to soil, collected from nitro aromatic contaminated site, could transform 1,3 dinitrobenzene (m-DNB) initially to m-nitroaniline, m-nitrophenol, m-aminophenol and resorcinol at 30 degrees C under shake culture condition. Carbon mineralization studies with unlabelled and radio labelled 1,3 (U14 C) dinitrobenzene subtrates indicated that the above metabolities appeared within 4 days. After 7 days incubation a significant traction of the source compound was degraded to C Q through aliphatic acids. Presence of nitro aryl reductase, aryl monooxygenase and resorcinol 1,3 di oxygenase was also noted in the sonicated cell mass.

ABSTRACT

Se presentan los resultados de la prueba de sensibilización al dinitroclorobenceno en un grupo de 103 pacientes con enfermedad de Hodgkin no tratados. Encontramos que fue posible sensibilizar al 80,6% de los pacientes, independientemente de la edad, el tipo histológico de la lesión, la extensión de la enfermedad y respuesta cutánea o antígenos de memoria convencionales. Sin embargo, la reactividad al dinitroclorobenceno apareció asociada a los conteos relativos de linfocitos con receptor para eritrocitos de carnero
